PCRの覚え書きPCR Protocol Notes

• 試薬を入れる順番：コンタミしたら困るものからチューブに入れる。 他の実験にも使う物を優先させること。 ただし、酵素は一番最後に入れる（失活したら困るから。）。基本的にMQ→buffer→dNTP→プライマー→鋳型→酵素の順番になると思います。Reagent addition order: Add reagents starting with those most sensitive to contamination. Prioritize shared reagents. Always add enzyme last (to prevent inactivation). Basic order: MQ → buffer → dNTPs → primers → template → enzyme.
• 酵素の種類：大きく3種類の酵素を使い分けています。Enzyme types: We use three main categories of enzymes.

• 1. 高正確：High fidelity: KOD-plus-Neo (TOYOBO)や or PrimeSTAR (TaKaRa)
• 2. 高PCR成功率：High amplification success rate: KOD FX Neo (TOYOBO)や or MightyAmp (TaKaRa)
• 3. 安い：Cost-effective: Taq polymerase (greiner)

• クローニングのためなら1.が良いでしょうし、なかなか増幅しないテンプレートなら2.が良いでしょう。大腸菌のインサートチェックは3.で充分です。For cloning use option 1; for difficult-to-amplify templates use option 2; for colony insert checks option 3 is sufficient.
