アガロースゲルの作り方How to Make Agarose Gels

1. アガロースゲル専用のプラスチック瓶に必要量のアガロース及び1xTAEを計り取る。Weigh the required amount of agarose and 1× TAE into a dedicated agarose gel plastic bottle.
• 1%; 1 g agarose/100 g 1xTAE, 2%; 2 g agarose/100 g 1xTAE, etc.
• アガロースゲル溶液の目安; 小1枚; 20 ml、大1枚; 40 mlApproximate volume: small gel 20 mL, large gel 40 mL
2. 電子レンジで3分程度（量による）加熱し、水蒸気の大きな気泡がでるまで加熱する。Microwave for approximately 3 minutes (depending on volume) until large steam bubbles appear.
3. スターラーで攪拌しつつ、手で触れるぐらいに冷めたら、100 mL当たり4μLのmidori greenを入れる。While stirring, allow to cool until touchable by hand, then add 4 µL of midori green per 100 mL.
4. アガロースゲル用トレイに流し込み、ピンセットでトレイの間の気泡を逃がす。Pour into the gel tray and remove air bubbles between tray walls with forceps.
5. コーム（バンド確認用は細いコーム、ゲルからの切り出し精製用は太いコームを使用する。）を差し込み、ラップを上にかけて平らな所で静置する（急いでいる時は冷蔵庫に入れても良い。）。Insert the comb (thin for band checking, wide for gel extraction/purification), cover with plastic wrap, and leave on a flat surface (can be refrigerated if in a hurry).
6. 固まったらコームを抜き、アガロースゲル用のタッパーに入れて冷蔵庫で保管する。Once solidified, remove the comb and store in an agarose gel container in the refrigerator.
